New GAG EndoNRE Biomarker Standards and Kits

We are introducing a new series of products based on the IDUA and I2S-associated GAG EndoNRE biomarkers.
The biomarkers previously designated UA-HNAc(1S)-Early and UA-HNAc(1S)-Late have now been structurally identified as IdoUA-GlcNAc6S and IdoUA2S-GlcNAc, respectively, through a collaboration with Professor Michael H. Gelb at the University of Washington. This work was supported by a National Institutes of Health (NIH) Small Business Technology Transfer (STTR) grant (1R41HD113422-01) and has been submitted for publication.
Building on this work, we have synthesized both biomarkers as qNMR-certified reference standards, together with their corresponding d3-labeled internal standards. This enables quantitative LC-MS/MS measurement using chemically identical stable-isotope-labeled internal standards for both biomarkers.
These standards are now available in various vial sizes on the Enfanos website.
This represents a significant analytical improvement over our GAG EndoNRE kits: BMKEG-1 through BMKEG-3. The previous kits used ∂UA-GalNAc4S as a chemically non-identical internal standard, providing semi-quantitative measurement of the two EndoNRE biomarkers. BMKEG-4 uses the d3-labeled internal standard for each biomarker, enabling quantitative LC-MS/MS measurement of IdoUA-GlcNAc6S and IdoUA2S-GlcNAc.
BMKEG-4-1000A and BMKEG-4-2500A are our next-generation kits for measurement of the GAG EndoNRE biomarkers associated with IDUA and I2S.
BMKEG-4 is designed as a duplex assay configuration. BMKEG-4-1000A provides reagents for approximately 1,000 assays and includes 2 nmol each of d3-IdoUA-GlcNAc6S and d3-IdoUA2S-GlcNAc. BMKEG-4-2500A provides reagents for approximately 2,500 assays and includes 5 nmol each of the two labeled internal standards. Both kits include PMP derivatization reagent supplied in 25-assay vials.
QC DBS materials are now sold separately to allow laboratories more flexibility in kit configuration. QCEG-5 and QCEG-6 are new 7-level and 3-level duplex DBS sets containing qNMR-certified IdoUA-GlcNAc6S and IdoUA2S-GlcNAc.
The 7-level and 3-level triplex DBS sets containing qNMR-certified IdoUA-GlcNAc6S, IdoUA2S-GlcNAc, and GlcNS-GlcUA (associated with SGSH) are also available.
PMP reagent and other components may be purchased in custom quantities, and custom kit configurations are available by quotation. BMKEG-1, BMKEG-2, and BMKEG-3 will remain available upon request. QCEG-1 and QCEG-2 will also remain available as custom-order products subject to minimum purchase requirements.
